• 1) Stability of SH3 Domain PI3K under varying conditions
• This involved the study of stability of the SH3 domain of PI3K under various concentrations of denaturant at different temperatures.The stability estimate of the protein is based on the analysis of denaturant induced equilibrium unfolding transitions measured spectroscopically using fluorescence as a probe at different temperatures. The measurements were extrapolated to standard conditions so as to obtain the values of ΔG, the free energy change and Cm, the mid-point of denaturation.
• 2) Preparation of the wildtype and single cysteine mutants SH3 Domain of PI3K
• This project included the expression, Isolation and Purification fo the said Proteins. The plasmid containing the required gene was transformed into BL21 DE3* cells and grown followed by induction with IPTG. The cells were then lysed and the protein was isolated with the help of Size exclusion chromatography. The protein thus obtained was then loaded onto an Ion-exchange Column followed by elution over a salt gradient using an HPLC. This protein was desalted with the help of a membrane with a cut-off of 3kDa using an Amicon.

• Histone deacetylases (HDACs) play an important role in gene expression and development by regulating chromatin modification. In an attempt to understand the importance of HDACs in development of Dicty (expand) from a biochemical point of view, I subcloned cDNA coding for two putative HDACs in E.coli expression vectors as GST- and His- fusion proteins. One of the two proteins could be induced but formed inclusion bodies in E.coli which was purified after urea solubilisation. The pure protein was used to inject in rabbits for raising an antiserum.
