Skills and Techniques: * Plasmid construction and transformation, vector engineering, High-throughput cloning techniques (SLIC,TA,Gibbson etc.),PCR and qRT-PCR variant, site-directed mutagenesis,PCR purification, primer designing and screening of clones in cost effective manner. * Cloning of ORFs and their expression, protein engineering, purification of recombinant proteins in high-throughput way using bacterial and animal cell systems. * Assay and method development for protein-DNA binding, Peptide modification and protein-protein interaction invivo and in-vitro system. * DNA-protein binding,aptamers specificity and affinity analysis using ChIP-seq, SELEX, DAP-seq. * Characterization and purification of proteins using techniques like analytical HPLC(C8 & C18),SDS-PAGE, Western Blot, dialysis, affinity,ion-exchange and size-exclusion chromatography (SEC). * Expertise in DNA and RNA analysis, DNA sequencing, RNA-seq, genomics and next generation sequencing (NGS) Miseq platform, Library preparation, Sonication, DNase treatment and purification. * Working expertise on handling biophysical techniques such as Ultracentrifugation(Beckman C.), Isothermal calorimetry (ITC), Thermal-shift assay (TSA), Circular Dichroism(CD) spectroscopy, UV-Vis Spec, EMSA (gel-shift assay), Fluorescence anisotropy, nano LC-MS/MS, MALDI-TOF, Mass spectrometry(ABsciex) analysis and Gel electrophoresis.
